>>116481. I actually think Glue would be a pretty funny name for it.
2. Keep essentially the same gameplay mechanics, just change the story and characters. There are six characters in the original game so the M6 work perfectly for this. Either assign roles based on color (as OP pic #2 does), or else you can slot them into roles based on personality:
>Twilight Sparkle: Professor PlumBecause purple and autism
>Rarity: Miss ScarletBest suited character to handle a femme-fatale role
>Applejack: Mr. GreenBecause Mr. Green is about money, and AJ is the only M6 pone who has a real job
>Rainbow Dash: Col. MustardOnly character who would make sense in a military role
>Fluttershy: Mrs. WhiteBecause Fluttershy would look good in a maid outfit, and also because natural submissiveness is a good quality for a servant to have
>Pinkie Pie: Mrs. PeacockBecause fuck it, it's the only one left. Mrs. Peacock is a widow who likes to party.
3. The new premise is that Princess Celestia has invited Twilight Sparkle and her friends to the Palace for some reason or other, but oh noes, she's been murdered. Since the culprit was obviously one of the six invited guests, the only sensible thing to do is to lock down the palace and have the six suspects investigate each other through a process of elimination. Makes way more sense than calling the ponice.
4. Gameboard can be identical, just make it pony-themed and Canterlot-themed. Most of the rooms would work just fine as locations in the palace.
5. Weapons could just be whatever. Change them to something pony-themed if you want: Giant cake (death by diabeetus), Party Cannon, whatever you want really. Or just keep the original weapons as they are.
6. I'm actually kind of liking this idea, and it seems like the kind of idea that could be banged out in a month. I think I might make a horror-themed murder mystery visual novel game loosely based on Clue mechanics, call it "Glue", and submit it for the /mlp/ game jam.
